If you are just starting the Dresden Files, I urge you to buy this book and read it as you go. Butcher includes a bit of a description at the beginning of each story so you know where they fit into the main timeline and you can read the mini-stories in between the main books.

If you are eagerly awaiting the release of Cold Days, I urge you to buy this book and use it to bide your time. Maybe go back and read through the series, plugging the shorts in between where they fit in.

There is not a lot of "missing" information in these stories - one answers the question of whatever happened to Jenny Greenteeth, but mostly they are stand alone features that were published in other anthologies and put into the hands of people who may or may not have been accustomed to Dresden's Chicago.
